1.动物解剖后, 尽快取材, 放入10倍自身体积of固定液中, 并选取合适of标本瓶, 充分固定24hours以上, 取材及固定时避免挤压, 造成人for损伤;
2.心肝脾肺肾, 肿瘤etc.常规tissue需用4%多聚甲醛固定后送样, 眼球, 脂肪, 睾丸etc.需用特殊固定液固定, 骨头需脱钙后进行实验;
3.送样时备注切片要求, 对位置have特殊要求of, such as脑. 请附带参考图;
4.若need白片, 请备注切片数量;

1.切片质量: 切片must完整, 均匀, 平整, 无气泡, 无褶皱, have利于stain时of冲洗and切片of牢固附黏. such as果切片不平整, 会导致stain不均匀, 颜色深浅不一.
2.固定and脱水: tissue固定要新鲜, 离体oftissue应尽快进行固定, 以防自溶and腐败. 固定时间and固定液浓度要适当, 避免tissue硬化and收缩. 脱水要彻底干净, tissue块 取材不能太大过厚, 以防脱水不完全.
3.stain前of准备: stain前应将切片in适当of缓冲液中浸泡, 调节pH值至适宜范围, 以防止tissue酸化或碱化影响cell核着色. 同时, 切片needthrough一系列of酒精浓度 逐渐脱水, 并用透明剂such as苯酚, 二甲苯etc.进行透明化, 以便于stain.
4.stain操作: stain时间长短needaccording to染剂对tissueofstain作用, 室温条件, 切片厚薄, 固定液of类别, 染液of新旧etc.因素进行调节. stain过程中要控制分色时间, generallyin显微镜下observecell核stain清楚而cell质基本无色for佳. 分化也is关键步骤, need掌握好分化时间and分化剂ofusing.
5.封片andobserve: 封片时要用中性树脂, 盖片要大于tissue块of面积, 以防日后褪色. observe时needin显微镜下observestain效果, such ashaveneed可进行调整.
